You can use both, it depends on the context. 'From' would be used in the context of using something that has some connection to the outside world.... 'work online from the comfort of your own home' but 'surf the internet in the comfort of your own home'. Basically 'from' implies some relationship with the outside world, and 'in' doesn't, although there are probably some exceptions.

Both make sense. I would argue that you would use 'from' if it is something that you would ordinarily have to travel for and for something more static. Nut, really, both are fine.

Either makes sense, i think the 'correct' phrase is from the comfort of your own home (or at least the one I personally have heard more often), but in reality you would get native speakers saying it both ways, i'm sure.
