The second one.
Although a day encompasses a period of time, when referred to in this specific way, you should use 'on'.
examples:
on thursday, I went to school
on new year's day, we celebrated
on my anniversary, I went to the city
but ...
during the evening ...
during the day ...
during the summer ...

Here you say :
"On my birthday....."
If you used 'during my birthday' it will rather be:
"During my birthday party .."
and you will mention events or incidents happening during the time your party took place as in :
"During my birthday party ,we danced and laughed a lot."
