Yes. It is technically correct. Preferred form, I have P.E. Tuesday afternoon.

Hi, Helen. You can not say 'of Tuesday' in English, it is grammatically incorrect. When talking about the day you did or will do something you must use 'on' You can say 'On Tuesday afternoon I have P.E. or I have P.E on Tuesday afternoon, or On Tuesday I have P.E in the afternoon.
