"And that is as it should be" means that whatever said before is good or makes sense in some way. So here it means means that death should be inescapable because the author sees it as the "single best invention in life." So "that" refers to death being inescapable. There are two "its" - both refer to death.

It means it is natural and right that death is inescapable. "That" refers "No one has ever escaped it.". "It" refers to the way things are, the natural way of things.
