The song is about love, the giving and receiving of love, the difficulties in the giving and receiving of love; and the beauty, wonder and grace of love. So that phrase in the song is about one who cannot receive love (be taken by love) and therefore cannot give love. A great song and a great movie.

Well, the line is a play on the opposites "give" and "take". "The one who won't be taken" can mean a person who refuses to be possessed and wishes to remain independent. However, "taken" can also mean "deceived" or "charmed/entranced". A person who is suspicious of other people's motivations cannot be generous. Or, a person who doesn't surrender him/herself emotionally has nothing to offer. There's no simple explanation here. Since it's a poetic line, it may be open to interpretation.

Not familiar with the song, but I'll take a guess. "Taken" is used in English to denote that someone is in a relationship. Example: "Oh, she's pretty. I'd like to go on a date with her." "Sorry, she's taken." If that's how it's being used in the song, it would be referring to someone who refuses to be in a relationship. They always remain single.
